Ammonia buildup is highly toxic

When your body breaks down protein, it makes a waste product called ammonia. Normally, this ammonia is turned into something called urea, which then leaves your body in your urine.
For people with urea cycle disorders (UCDs), this process doesn’t work correctly, and ammonia can build up in the blood.
Too much ammonia is bad for the brain. It can make it hard to think, remember, learn, or act like your usual self. Uncontrolled ammonia can cause dangerous health problems and hyperammonemic crises (HACs).*
OLPRUVA is for the treatment of certain UCDs and not for the treatment of acute hyperammonemia. This condition is a medical emergency caused by rapidly rising, uncontrolled ammonia levels that can be life-threatening and requires immediate medical treatment.

How to spot uncontrolled ammonia

UCD symptoms present differently by age of onset

Symptoms in older individuals can be more subtle and are often triggered by excessive exercise, illness, or high-protein meals. Here are some, but not all, of the most common symptoms in older children and adults:

Neurological Symptoms:

Lethargy/fatigue, confusion, agitation, sleep disorders, behavioral changes, psychiatric symptoms (delusions, hallucinations, psychosis), and stroke-like symptoms.

Gastrointestinal Symptoms:

Nausea, vomiting, and loss of appetite.
While UCDs can affect individuals from infancy to adulthood, OLPRUVA is only prescribed to those 1 year old or older, weighing 15 pounds (7 kg) or greater.

Taking essential amino acid supplements

An important part of managing UCDs is to work with your healthcare team to establish a treatment plan to monitor and maintain ammonia control in order to maximize physical and mental functioning.
Certain medicines may increase the level of ammonia in your blood or cause serious side effects when taken during treatment with OLPRUVA. Tell your healthcare provider if you or your loved one takes corticosteroids, valproic acid, haloperidol, or probenecid.
